Research Summary:

Natural products (NPs) are of great pharmaceutical importance and play an indispensable role in modern drug development as NPs or their derivatives comprise over half of modern small molecule drugs. However, in recent decades, there has been a decline in NP-based drug development. This is in part due to a number of challenges associated with decreased frequency of identifying novel NP structures with traditional methods, obtaining sufficient supply of NP lead compounds through the drug development pipeline, and modifying the highly complex NP structures for lead optimization. Recently, the leveraging of genome sequence information has allowed greater access to NP biosynthetic pathways. Advances in synthetic biology are enabling these complex biosynthesis schemes to be efficiently reconstructed in fermentable microbial hosts using novel genetic and molecular tools. Such synthetic biosynthesis platforms offer an efficient and versatile alternative to direct isolating compounds from natural producers and/or chemical synthesis for NP sourcing and derivatization.

We are specifically interested in:

1) Harnessing synthetic biology to more efficiently understand the biosynthesis of complex natural products.

2) Developing synthetic biology tools to enhance the bio-production of costly natural products to commercial production level.

3) Discovery of novel natural products to advance natural product-based drug discovery through engineering approaches.
