Protein and Lipid Microarrays with Labeled (Fluorescence) and Label-free (Surface Plasmon Resonance Imaging) DetectionResearch Summary:
Surface plasmon resonance (SPR) is a surface-sensitive analytical technique that measures the change in refractive index of a molecular layer adjacent to a metal film. A surface plasmon can be regarded as a bound evanescent wave propagating along the metal-dielectric interface. The electric field decays exponentially from the surface (~200 nm), providing high measuring sensitivity to changes in its vicinity. SPR is widely used in biomolecular interaction analysis (BIA), and offers a variety of functions, including real time detection and kinetic analysis, without the need of a label. Our lab studies protein interactions with microarray technology in combination of SPR imaging detection. One of the current focus areas is lipid microarrays. Compared to DNA microarray (mature and commercially successful) and protein microarray (rapidly improving), lipid microarray is a relatively new development. Recent progress in supported membranes has provided new tools to generate membrane arrays and templates. Our latest work on calcinated silicate films on gold also offers a novel surface for the development of the lipid microarray/SPR imaging technology.
